Senescence

Research suggests that FOXO4-DRI may increase the regeneration of cells by inducing apoptosis in senescent cells. A 2017 study on aged mice models reported that those given the peptide appeared to show signs of improved fitness, better renal function, and increased fur density. This is believed to be related to the peptide's potential to reduce the burden of senescent cells, which contribute to aging through a pro-inflammatory environment.

Cardiovascular Function & Insulin Signaling

Research suggests that proteasome enzymes, which are responsible for removing dysfunctional cells, decrease over time. The naturally occurring FOXO4 protein appears to regulate these enzymes, but FOXO4-DRI is being investigated for its potential to not only regulate this process but also aid in the elimination of dysfunctional cells.

FOXO4's activities are also often linked to the insulin and insulin-like growth factor signaling (IIS) pathway. This pathway influences how cells respond to stress and growth signals, and FOXO4 is believed to mediate the effects of diet and environmental factors on cellular aging.


Hypogonadism

A study on senescent Leydig cells (crucial for testosterone production) suggested that FOXO4-DRI may selectively eliminate these aging cells. The peptide appeared to prevent FOXO4 from binding to p53, allowing p53 to induce the apoptosis of the senescent Leydig cells. This suggests that the peptide may help alleviate some of the dysfunctions associated with aged Leydig cells, such as reduced testosterone synthesis.
