Research and Clinical Studies
Growth Hormone Secretion & Synergistic Potential
Both Mod GRF 1-29 and GHRP-2 are being investigated for their potential to stimulate growth hormone release from the pituitary gland. Mod GRF 1-29 appears to activate the adenylyl cyclase pathway, which leads to an influx of calcium ions into the cells and the release of GH (11). GHRP-2 is believed to activate ghrelin receptors, also triggering GH release (5).
Research suggests that when combined, these two peptides may have a synergistic effect on GH secretion. For example, while one study found that GHRP-2 alone may lead to a 47-fold increase in pulsatile GH secretion, and a GHRH-mimetic may lead to a 20-fold increase, their combination appeared to induce a 54-fold increase in pulsatile GH secretion, which suggests a synergistic potential (11).
Appetite & Metabolism
Due to the presence of GHRP-2, which activates ghrelin receptors, the peptide blend is being investigated for its potential to increase appetite. A clinical trial on test subjects reported that the peptide group consumed about 35% more food than the placebo group, with each subject exhibiting increased appetite (6).
A literature review of studies on GHSs suggests that these peptides may also lead to an increase in lean body mass, a reduction in fat mass, and an improvement in sleep cycle in mature and obese subjects (7).
Tolerability & Hormone Levels
Animal studies have been conducted to evaluate the general effects of these peptides. In guinea pigs and rabbits, the peptides did not appear to have any profound effects on organ systems, with only minor reported changes in intestinal motility. Researchers have commented that the peptide may be well tolerated and potentially useful for diagnosing or treating GH deficiency (5, 9).
A clinical study on both younger and older male subjects showed that while GH levels increased in both groups after exposure to GHRP-2, the increase was more significant in the younger group. The peptide also appeared to stimulate increased levels of ACTH and cortisol, as well as a mild increase in prolactin (10).
