Selective Targeting: The peptide is hypothesized to selectively target cancer cells because it binds specifically to the HDM-2 protein, which is overly expressed on the membranes of cancerous cells but not typically found on the membranes of healthy cells.
Membrane Perforation: The binding of PNC-27 to HDM-2 is thought to lead to the formation of transmembrane pores in the cancer cell membrane.
Necrosis: The formation of these pores causes a physical disruption of the cell membrane, allowing the influx of ions and the efflux of cellular contents. This process, known as membranolysis, ultimately leads to cell death via necrosis rather than apoptosis (programmed cell death).

Cancer Cell Lysis
Selective Cell Death: A 2009 study suggested that PNC-27 appears to form stable complexes with HDM-2, leading to the formation of ring-shaped pore structures only in cancer cells. These structures were not observed in normal cells.
Intact Peptide Action: Research using fluorescent labeling showed that PNC-27 remained intact during the process of membranolysis, suggesting that its full structure is crucial for its activity.
Non-Solid Tumors: A 2014 study on non-solid tissue tumor cells (leukemia cell lines) suggested that HDM-2 was also highly expressed on their membranes and that PNC-27 exhibited selective toxicity toward these cells, inducing cell death in a similar manner to its action on solid tumors.
Tumor Growth: A study on a similar peptide, PNC-28, suggested that it may inhibit the growth of ovarian cancer cells and reduce tumor size in mouse xenograft models.
